Analysis of the interaction between sulfonated sugars and Alzheimer Aβ

Abstract

Biological active saccharide of proteoglycans and gangliosides has been reported to accelarate the amyloid formation of Alzhemir peptide. Those reports indicated the strong interaction with Ab. We have reported the synthesis and biological activity of sulfonated sugars for a sialic acid mime. Here, we investigate the interaction between sulfonated GlcNAc and Aβ42. The aggregation of Aβ with heparin and pNP 6-sulfo-GlcNAc was estimated by fluorescence of thioflavin T, CD spectra and TEM observations. Though heparin induced the aggregation and β-sheet confomation, pNP 6-sulfo-GlcNAc inhibited them. Other saccharides like glucose and pNP GlcNAc did not show the inhibition effect.
