Atto- and femto-litre microfabrcated structures for investigating depression of enzyme activity at a single molecule reaction

Abstract

In this work, we successfully measured a single enzyme reaction in a microfabricated chamber -(510 aL, 7.2 fL, 61 fL and 624 fL) and estimated the effect of enzyme adsorption and denaturation on the reaction rate with inner-surface coated and non-coated chambers. Our results indicate that a single enzyme reaction rate decreased with decreasing the chamber size and that reaction rate in coating chambers slightly increased compared to that in non-coated one.
