Biofunctional characteristics of dendritic glycocluster modified surfaces

Abstract

Oligosaccharides on cell membranes are known to interact with exterior biomolecules, and thus they play important roles in living systems. The interaction between oligosaccharides and such biomolecules is usually weak, but is often strengthened by the clustering of oligosaccharides. This elevation phenomenon is known as the "glycocluster effect", and many glycomaterials had been developed, based on this idea. Glycodendrimers which have a definite molecular weight and structure are one type of such glycocluster. We had already synthesized various glycodendrimers and evaluated their interaction with lectins, and also prepared functional glycodendrimer layers. In this paper, we utilized dendritic glycoclusters that were immobilized on surfaces for the detailed analysis of the interaction between the saccharides and lectins or other proteins such as amyloid β. Based on the kinetic analysis, we can evaluate the potency of the surface for molecular-binding.
