Detection of fungicide-resistant pathogens by weak-affinity capillary electrophoresis using DMA-PEG conjugates

Abstract

We developed a method for separating a mixture of DNAs having a single-base difference by using weak affinity capillary electrophoresis (WACE). DNA-poly(ethylene glycol) (PEG) conjugate was synthesized as a separation medium of WACE. A DNA unit of the conjugate was designed to be complementary to the wild-type sample DNA and was coupled with a PEG unit for retardation of the electrophoretic migration in the capillary to act as an affinity-probe against wild-type sample DNA. The DNA-PEG conjugate properly worked as a separation medium, and a mixture of 60 mer wild-type DNA and its single-base mutant was successfully separated by the present method.
