Development of a hybrid artificial liver using multi capillary PUF/Spheroid packed bed

Abstract

We have already reported that rat hepatocytes/spheroid culture using polyurethane foam (PUF) as a culture substratum is a useful method for keeping of the hepatic functions and designing of a hybrid artificial liver module. In this study, Multi Capillary (MC) PUF packed-bed culture system was developed to downsize a hybrid artificial liver module for medical care use. This module has the high PUF packing ratio (79%) and the uniform channels for medium flow, and is easy to scaling up. We also developed the inoculation method of hepatocytes to MC-PUF using centrifugation (centrifugation method). A high cell immobilizing ratio (maximum value was 80%) and a high cell density (maximum value was 1.8 x 107 cells/cm3-PUF) in MC-PUF packed-bed was achieved by this method. We could also observe the spheroid formation of hepatocyte in MC-PUF under less than 0.79 x 107 cells/cm3-PUF of cell density. From the estimated performace of this MC-PUF/spheroid packed-bed culture system, the required vessel volume for immobilizing 1/4 hepatocyte number of adult human liver is calculated to be 5 L, and the required vessel volume for substituting 1/4 liver function (urea synthesis) of adult human is about 18 L. These values as a hybrid artificial liver are much smaller (about 1/10) than that of our previously designed vessel. These results indicate that this system seems to be available for medical care of hepatic failure.
