Microheater-nanowires device for detection of cell-free circulating methylated DNA

Abstract

Detecting of cell-free circulating methylated DNA could be a promising method for early diagnosis of cancer. Here we have developed the microheater - zinc oxide nanowires (ZnO-NWs) structures embedded in the microchannel device for capturing DNA via electrostatic interaction. We found that 60% of DNA could be captured on ZnO-NWs. Additionally, the microheater, which constantly enables to generate heat, can denature double stranded DNA (dsDNA) to single stranded DNA (ssDNA). Finally, ICON Probe, which is an artificial ssDNA probe for DNA methylation, was introduced inside a device to bind to ssDNA for DNA methylation detection.
