Photo-enhanced efficiency of cationic lipid-mediated transfection using cationic azobenzene-based lipid

Abstract

Self-assembling behavior of cationic azobenzene-based lipid (KAON12) was directly examined by transmission electron microscopy and phase-contrast microscopy with small unilamellar vesicles or giant liposomes. In a transfection experimenta using the cationic lipid, UV irradiation improved the transfection efficiency of KAON12 compared without photoirradiation. After KAON12/DNA complex passes through the plasma membrane by endocytosis, at endocytic vehicles, trans to cis photo-isomerization of azobenzene moiety would destabilize the vehicle membrane, thus accelerating membrane fusion of photo-responsive lipid vesicles. This phenomenon aids the escape of plasmid DNA from endocytic vehicles, thus improving transfection efficiency with UV irradiation.
