Selective incorporation of hydrophobic probe into schzophyllan(SPG)

Abstract

β-(1-3)-D-glucan schizophyllan (SPG) can form a triple helix and single random coil(s-SPG) in a neutral pH and basic (pH>13) water, respectively. It is known that s-SPG could interact with some hydrophobic molecules. In this report, we examined interaction of SPG and hydrophobic molecule using 2,6 ANS as a hydrophobic probe. When 2,6 ANS was added to s-SPG, blue-shifted fluorescence spectrum was observed and the intensity was also increased. Furthermore, the mixture of 2.6-ANS and s-SPG was observed induced-CD band at 328nm in CD spectrum. In the presence of dextran, amylose and triple-stranded SPG, these spectral changes were not observed. These results suggest that 2.6-ANS bound to s-SPG with stacking interaction.
