A microfluidic molecular tracking system detects a time-to-search DNA sequence to be cleaved by restriction enzymes

A microfluidic analytical system, which uses simple technique to pick reacting DNA molecules out of a crowd of molecules during enzymatic reaction, was developed on the basis of single-molecule imaging. Restriction enzymes EcoR I and Not I with their target and non-target DNA were analyzed. With this system, we measured the duration difference of restriction-site-searching by these enzymes at the single-molecule level.
