Primary dog hepatocytes formed spheroid in the pore of polyurethane foam (PUF) spontaneously. Then, we tried to scale up multi-capillary PUF/spheroid packed bed module (PUF volume; 300cm 3) as a hybrid artificial liver using dog hepatocytes. These hepatocytes also formed spheroid in this module and maintained the almost same activity of albumin secretion as that in stationary culture. Furthermore, we developed extracorporeal circulation system including this artificial liver module and estimate the performance of the module using warm ischemic liver failure dog. Consequently, blood glucose concentration was maintained, and increases of blood ammonia and LDH were suppressed by artificial liver treatment. These performances of this module were comparable or slightly superior than those of Demetriou's artificial liver module which was already used clinically as a bridge use for liver transplantation. These results indicate that the PUF/spheroid packed-bed module seems to be promising as a hybrid artificial liver.
