N2 - We developed a new method to isolate a natural pigment of mesobiliverdin IXα (MBV) directly from phycocyanin of Thermoleptolyngbya sp. O-77 and synthesized a water-soluble Cp*Ir complex with the isolated MBV ligand. To obtain a high yield of MBV, we modified the conventional method by adding additional two steps: the protease digestion of phycocyanin by trypsin and the isolation of MBV by hydrophobic and silica gel column chromatographies. The Cp*Ir complex with MBV ligand was characterized by electrospray ionization-mass spectrometry, ultraviolet-visible-near-infrared absorption spectroscopy, X-ray photoelectron spectroscopy, 1H NMR spectroscopy, and H–H correlation spectroscopy. This is the first example of a platinum-group metal complex with MBV as a ligand.
