We recently proposed the novel gene-regulation system responding to cAMP-dependent protein kinase (PKA). For this system, we used the cationic polymer which includes the oligopeptide that is the substrate of PKA. Since this polymer forms complexes with DNA, the gene expression was totally suppressed. However, when PKA is being continuously activated, anions were inserted in the polymer by phospholilating the peptides of side chains, the complex of DNA and polymer collapses and transcription of the gene is activated. We attempted to apply this system to living cell, and were successful to intracellular target signal-responsive regulation of delivered gene-expression.
