There is a need for a pancreatic lipase (LIP) reference assay to provide a clinical usefulness base to which routine methods can be traceable. The method proposed herein as a candidate reference method is a coupled enzymatic method using 1,2-dioleoylglycerol as the substrate. The reaction is spectrophotometrically monitored by measuring formation of NADPH, and the constituent chemicals for the reaction are all commercially available, which makes it feasible for this assay to be performed in any laboratory. With this method, LIP activity was linear up to 440 U/L (eight times expected upper limit of physiological concentration) . The between-laboratory variation for six samples surveyed at five laboratories was 3.80% to 26.4% (CV) for samples containing about 20 to 290 U/L LIP activity. Good correlations were obtained with five routine kits. The method may be likely applicable as one candidate reference method. We report here the optimization, properties and the transferability of the method.
