Effect of exogeneous 5-aminolevulinic acid on fungal production of extracellular heme peroxidases

The effect of exogenous 5-aminolevulinic acid (ALA) on the production of extracellular heme peroxidases was investigated. The addition of ALA (2.0 mM) to white-rot fungi cultures increased lignin and manganese peroxidases (LiP and MnP) activities 5- and 10-fold, respectively, relative to the conventional ligninolytic cultures. However, the exogenous glycine and glutamate, biosynthetic precursors of ALA had no effect on LiP and MnP activities. It was also found that ALA did not act as a stimulator for LiP and MnP. A quantitative analysis revealed that ALA increased intracellular concentration of free heme. These results suggested that the rate determining step for fungal production of extracellular heme peroxidases is most likely ALA biosynthetic pathway.
