Efficient detection of protein-protein interactions on PMMA stereocomplex films

Isotactic and syndiotactic poly(methyl methacrylate)s (PMMAs) form double-stranded helical stereocomplexes with a respective molar stoichiometry of 1:2 based on van der Waals interactions in organic solvents. Bioactivities of stereocomplex surfaces are significant for determining the potential for biomedical applications. We prepared PMMA stereocomplex films by a layer-by-layer assembly method, and the films were coated with protein A. Antibodies were deposited on the films, and then the binding of antigen was analyzed by QCM, AFM, and ATR-IR measurements. Specific binding amounts of HSA on Anti-HSA-coated stereocomplex films were greater than other surfaces and activity of the immobilized protein A on stereocomplex films was maintained for 10 days. These observations indicate that the surfaces of stereocomplex films were superior to conventional films for analysis of protein-protein interactions.
