Genetical studies on the “non-molting k” mutant in the silkworm, Bombyx mori

Larvae whose growth was retarded were found in one batch of a cross performed between the strains e34 and w23 maintained in Kyushu University. These retarded individuals, about one fifth of the batch, could not enter molt and died. On the basis of genetical studies, this character was ascribed to a mutation of a single recessive gene. Thus the mutant was named “non-molting k” with the gene symbol nm-k. Linkage analysis showed that nm-k was linked with L and sk which had been mapped at the 15.3 and 41.1 positions, respectively, of the 4th linkage group. The nm-k gene was precisely localized by a three-point experiment involving L, sk and nm-k. The recombination value between L and nm-k was calculated to be 14.16%, between nm-k and sk 17.66% and between L and sk 31.09%. Hence the arrangement of these 3 gene loci on the chromosome is in the order of L-nm-k-sk. Taking a correction factor into account, the locus of nm-k was determined to be 26.8 of the 4th linkage group.
