Propagation of plantlets by the culture of multiple shoots and roots induced from callus originating in the cambium of Robinia hispida L

A culture system suitable for the regeneration of plantlets by the culture of multiple shoots and roots induced from callus originating in branch cambium of a two-year-old graft of a rose acacia (Robinia hispida L.) mature tree was tested and established. Inducement of callus from the cambium was very easy using medium supplemented with 1.0 mg/l of 2, 4-D. Adventitious buds were differentiated from the callus as a result of the culture by liquid 1/2 Ms (Murashige-Skoog) medium supplemented with 1.0 mg// of IAA (indole-3-acetic acid) of 0.1 mg// BAP (6-benzylaminopurine) in a reciprocal shaking shaker. These buds induced multiple shoots and shoot growth by culture on solid 1/2 MS medium supplemented with 0.1 mg/l of BAP. Rooting from these shoots was observed on vermiculite moistened with sterile distilled water. On the other hand, adventitious roots were differentiated from calli originating in the cambium. The roots were kept propagating by culture with a reciprocal shaking shaker in a dark condition on liquid 1/2 MS medium supplemented with 1.0 mg/l of IAA. Adventitious buds and plantlets were induced through callus formation from these roots.
