We designed gene carriers that can activate the delivered gene in response to the intercellular kinase signal. 'c-Src', which is a non-receptor tyrosine kinase, is specifically activated within a cancer cell. Our goal is to create various gene carriers to express a therapeutic gene in response to 'c-Src' activity. First, we synthesized some polymers that are grafted with c-Src substrate peptides as an intercellular signal-response gene carrier. Then, we evaluated whether those carriers can regulate gene expression in cell free system. We will discuss the relation between the carrier design and their abilities for signal specific gene regulation.
