The hierarchical structure of a hollow callose fiber secreted from protoplasts

A gigantic fiber was secreted from a protoplast under the stressful condition. It was clarified that the fiber was made of β-1,3-glucan (callose) using histochemical methods. The results of microscopy indicated that the fiber owned hierarchical structure composed of hollow sub-fibrils. We also attempted to obtain building units comprising the callose fiber. The individual units were in several sizes and forms, such as a sheet, a micro fiber, triple and single helical chains, obtained from the fiber by mechanical and chemical methods. This indicates that the secreted callose fiber was assembled by hollow sub-fibrils also composed of microfibrils that comprise triple and single helical chains of β-1,3-glucan molecules.
